Osmosis and pervaporation in polyimide submicron microfluidic channel structures

    Get PDF
Osmosis and pervaporation of water through the roof of all-polyimide channels of 500 nm height is described. The phenomena cause both a liquid flow in the channels and a concentration change of dissolved salt. Both effects are amplified due to the thin channel roof and the small channel height. Osmotic movement of demineralized water was observed towards a salt solution and towards ethanol and isopropanol. Water movement by pervaporation was observed from a salt solution towards the atmosphere. Flow velocities of up to 70 ?m/s were generated in the channels. The results are in accordance with predictions from the solution-diffusion model for membrane transport. The observed phenomena can be applied in a nanofluidic osmotic pump or for an osmotic or pervaporative concentrator

Large scale patterning of hydrogel microarrays using capillary pinning

Capillary barriers provide a simple and elegant means for autonomous fluid-flow control in microfluidic systems. In this work, we report on the fabrication of periodic hydrogel microarrays in closed microfluidic systems using non-fluorescent capillary barriers. This design strategy enables the fabrication of picoliter-volume patterns of photopolymerized and thermo-gelling hydrogels without any defects and distortions

Evaluating deterministic motif significance measures in protein databases

Abstract

Background

Assessing the outcome of motif mining algorithms is an essential task, as the number of reported motifs can be very large. Significance measures play a central role in automatically ranking those motifs, and therefore alleviating the analysis work. Spotting the most interesting and relevant motifs is then dependent on the choice of the right measures. The combined use of several measures may provide more robust results. However caution has to be taken in order to avoid spurious evaluations.

Results

From the set of conducted experiments, it was verified that several of the selected significance measures show a very similar behavior in a wide range of situations therefore providing redundant information. Some measures have proved to be more appropriate to rank highly conserved motifs, while others are more appropriate for weakly conserved ones. Support appears as a very important feature to be considered for correct motif ranking. We observed that not all the measures are suitable for situations with poorly balanced class information, like for instance, when positive data is significantly less than negative data. Finally, a visualization scheme was proposed that, when several measures are applied, enables an easy identification of high scoring motifs.

Conclusion

In this work we have surveyed and categorized 14 significance measures for pattern evaluation. Their ability to rank three types of deterministic motifs was evaluated. Measures were applied in different testing conditions, where relations were identified. This study provides some pertinent insights on the choice of the right set of significance measures for the evaluation of deterministic motifs extracted from protein databases.</p

Hypertension Is Associated with Marked Alterations in Sphingolipid Biology: A Potential Role for Ceramide

    Get PDF
Background Hypertension is, amongst others, characterized by endothelial dysfunction and vascular remodeling. As sphingolipids have been implicated in both the regulation of vascular contractility and growth, we investigated whether sphingolipid biology is altered in hypertension and whether this is reflected in altered vascular function. Methods and Findings In isolated carotid arteries from spontaneously hypertensive rats (SHR) and normotensive Wistar-Kyoto (WKY) rats, shifting the ceramide/S1P ratio towards ceramide dominance by administration of a sphingosine kinase inhibitor (dimethylsphingosine) or exogenous application of sphingomyelinase, induced marked endothelium-dependent contractions in SHR vessels (DMS: 1.4±0.4 and SMase: 2.1±0.1 mN/mm; n = 10), that were virtually absent in WKY vessels (DMS: 0.0±0.0 and SMase: 0.6±0.1 mN/mm; n = 9, p Conclusions Hypertension is associated with marked alterations in vascular sphingolipid biology such as elevated ceramide levels and signaling, that contribute to increased vascular tone

A Proposal For Fuzzy Rule Generation Using Temporal Reasoning

Knowledge acquisition is difficult, especially when the domain knowledge is not structured. In this paper a framework based on machine learning is proposed in order to generate a rule-base for signal analysis in the case of anesthesia monitoring. During surgery, clinical parameters are measured. From training samples, rules for a knowledge-based system can be learned that describe alarm situations in terms of the clinical parameters. The training samples consist of time series of the clinical parameters and an expert decision which defines an alarm or noalarm situation. The samples are classified into concepts. Fuzzy rules are generated that relate the concepts with a decision. By means of temporal reasoning on the time series, fuzzy temporal concepts and rules can be generated. The (temporal) concepts are used to recognize an alarm situation. The framework can be applied to image sequence analysis as well. 1. Rapid sacrificial layer etching for the fabrication of nanochannels with integrated metal electrodes

    Get PDF
We present a rapid etch method to surface-micromachine nanochannels with integrated noble metal electrodes using a single metal sacrificial layer. The method is based on the galvanic coupling of a chromium sacrificial layer with gold electrodes, which results in a 10-fold increase in etch rate with respect to conventional single metal etching. The etch process is investigated and characterized by optical and electrochemical measurements, leading to a theoretical explanation of the observed etch rate based on mass transport. Using this explanation we derive some generic design rules for nanochannel fabrication employing sacrificial metal etching

Continuous flow microfluidic demixing of electrolytes by induced charge electrokinetics in structured electrode arrays

    Get PDF
A continuous flow microfluidic demixing process is realized. It utilizes high external electrical fields that are applied over electrically floating noble metal electrodes in an otherwise straight microchannel. The process converts axial electrical potential gradients into lateral molecular selective transport via a structure oriented ensemble of numerous electrodes. While the individual electrodes locally modify the electrolyte distribution by nonlinear electrokinetic effects and concentration polarization, the directed orientation of the electrode array combines the individual polarization zones to a dedicated molecular enrichment against the generated concentration gradient. A homogeneously concentrated electrolyte can be separated into arbitrarily shaped laminae of increased and depleted concentration by the presented microfluidic demixer. \u
